SPH3127 + valsartan is a combination of two antihypertensive agents. SPH3127 is an oral, non-peptide, small molecule direct renin inhibitor developed by Shanghai Pharmaceuticals Holding and originally by Mitsubishi Tanabe Pharma Corporation. It acts by inhibiting renin, the rate-limiting enzyme in the renin-angiotensin-aldosterone system (RAAS), thereby reducing blood pressure and providing renal protection. Valsartan is an angiotensin II receptor blocker (ARB) that lowers blood pressure by blocking angiotensin II from binding to its angiotensin II receptor type 1 (AT1R) on vascular smooth muscle and adrenal gland tissue, leading to vasodilation and reduced aldosterone secretion. The combination targets two different points in the RAAS pathway for enhanced antihypertensive efficacy. This combination has been studied in phase 3 clinical trials for hypertension[1][8][6].
