SPX-303 + trastuzumab deruxtecan is a combination therapy being developed by SparX Biotech. It consists of SPX-303, a bispecific antibody targeting Leukocyte Immunoglobulin-Like Receptor B2 (LILRB2) and Programmed Death-Ligand 1 (PD-L1), and trastuzumab deruxtecan (Enhertu), a HER2-directed antibody-drug conjugate (ADC). SPX-303 is designed to reprogram the immunosuppressive tumor microenvironment by blocking LILRB2 on myeloid cells and inhibiting the PD-1/PD-L1 checkpoint. Trastuzumab deruxtecan provides targeted delivery of a topoisomerase I inhibitor payload to HER2-expressing tumor cells. The combination is currently in Phase 1 clinical trials for the treatment of solid tumors, aiming to leverage the synergistic effects of immune checkpoint inhibition and ADC-mediated cytotoxicity.
