SPY001 + SPY002 is an investigational combination therapy comprised of two extended half-life monoclonal antibodies developed by Spyre Therapeutics for the treatment of inflammatory bowel disease (IBD), especially ulcerative colitis and Crohn’s disease, and potentially rheumatoid arthritis and other immune-mediated conditions. SPY001 targets the integrin α4β7 to block its interaction with MAdCAM-1, thus limiting the migration of inflammatory immune cells to the gut and reducing local inflammation. SPY002 targets the cytokine TL1A, blocking its binding to death receptor 3 (DR3) on immune cells and inhibiting pro-inflammatory signaling pathways. Both antibodies are engineered for infrequent subcutaneous or intravenous dosing; SPY001 mimics and potentially improves upon the biology of vedolizumab, while SPY002 has shown a prolonged half-life and potent TL1A suppression in early-phase studies. Preclinical and early clinical results suggest the combination of these two agents may produce additive or synergistic effects by orthogonally targeting two relevant and complementary pathways in IBD and other immune diseases[1][3][6].
