SPY001 + SPY002 + SPY003 is a combination of three investigational extended half-life monoclonal antibodies, each targeting a distinct, clinically validated mechanism in inflammatory bowel disease (IBD): α4β7 integrin (SPY001), TL1A cytokine (SPY002), and interleukin-23 (IL-23, SPY003). This combination is developed for the treatment of ulcerative colitis and potentially Crohn’s disease, aiming to deliver synergistic efficacy beyond monotherapy by simultaneously suppressing intestinal lymphocyte trafficking, pro-inflammatory cytokine signaling, and Th17-mediated immune activation. Spyre Therapeutics is advancing this combination into Phase 2 platform trials. All antibodies are optimized for convenient quarterly or biannual subcutaneous administration with improved pharmacokinetics over current standards of care. Early clinical and preclinical studies show favorable safety, tolerability, and no drug-drug pharmacokinetic interactions among the components[1][2][3][5][6][13][15].
