SQZ-eAPC-HPV + pembrolizumab is a combination of a novel mRNA-engineered autologous cell therapy, derived from peripheral blood mononuclear cells (PBMCs) and engineered to target the HPV16 E6 and E7 oncoproteins, with the immune checkpoint inhibitor pembrolizumab. SQZ-eAPC-HPV utilizes the Enhanced Antigen Presenting Cell (eAPC) platform, delivering multiple mRNAs encoding tumor antigens, a costimulatory molecule (CD86), and membrane-bound cytokines (IL-2 and IL-12) into four types of immune cells (monocytes, T cells, B cells, NK cells). Its mechanism is to stimulate antigen-specific T cell responses against HPV16+ tumor cells. Pembrolizumab acts by blocking programmed death-1 (PD-1) receptor, preventing immune suppression. This combination aims to enhance immune recognition and killing of HPV16+ solid tumors, and is being evaluated in a Phase 1/2 clinical trial (COMMANDER-001) for patients with recurrent, locally advanced, or metastatic HPV16+ solid tumors, including head and neck, cervical, anal, vulvar, or penile cancers[1][3][4][5][6][7].
