Overview

SQZ-PBMC-HPV + nivolumab is a combination investigational immunotherapy regimen for HPV16-positive advanced or metastatic solid tumors. SQZ-PBMC-HPV is an autologous cell therapy product developed using SQZ Biotechnologies’ Cell Squeeze technology to engineer peripheral blood mononuclear cells (PBMCs) ex vivo to present HPV16 antigens, aiming to stimulate a tumor-specific CD8+ T cell response. Nivolumab is a monoclonal antibody checkpoint inhibitor targeting programmed cell death protein 1 (PD-1), enhancing immune system anti-tumor activity by blocking immune inhibitory signaling. The combination seeks to augment antigen-specific T cell activation by both directly priming T cells via the cell therapy and reducing immunosuppressive signaling via PD-1 blockade. The main clinical focus is HPV16-positive recurrent/metastatic cancers, including head and neck, anal, and cervical cancers[1][4].
