SR-8541A + nivolumab is a **combination therapy** consisting of SR-8541A, a potent, orally available small molecule inhibitor of the enzyme **Ectonucleotide pyrophosphatase/phosphodiesterase 1 (ENPP1)**, and **nivolumab**, a fully human **monoclonal IgG4 antibody** that acts as an **immune checkpoint inhibitor** targeting **programmed cell death protein 1 (PD-1)**. SR-8541A, developed by Stingray Therapeutics, promotes STING pathway activation, stimulates type I interferon and chemokine expression, and augments anti-tumor immunity by increasing immune cell infiltration and modulating the tumor microenvironment. Nivolumab, primarily developed by Bristol Myers Squibb, blocks PD-1 interaction with its ligands (PD-L1, PD-L2) to restore T-cell activity and enhance immune-mediated anti-tumor responses. The combination is being studied in advanced/metastatic solid tumors and is expected to synergistically enhance immune activation, particularly in tumor types where monotherapy checkpoint inhibition is ineffective or resistance has developed.
