SRK-181 + anti-PD-(L)1 antibody therapy is an investigational combination drug regimen comprised of SRK-181, a selective monoclonal antibody inhibitor of latent transforming growth factor-beta 1 (TGFβ1) activation, and an anti-PD-(L)1 immune checkpoint inhibitor antibody. SRK-181 specifically binds and inhibits the activation of latent TGFβ1 without significant activity on TGFβ2 or TGFβ3, aiming to overcome tumor microenvironment-mediated resistance to PD-1/PD-L1 blockade. Anti-PD-(L)1 antibodies are designed to enhance anti-tumor immunity by blocking programmed cell death protein 1 (PD-1) or its ligand PD-L1. The combination is investigated primarily in patients with locally advanced or metastatic solid tumors who are non-responders to prior anti-PD-(L)1 monotherapy, with the goal of enhancing response rates to immune checkpoint blockade in resistant cancers[1][2][3][4][5].
