Drug intelligence / Profile preview
SS1P + cisplatin + pemetrexed is an investigational combination therapy for cancer, particularly mesothelioma. The regimen combines three agents: - **SS1P** is a recombinant immunotoxin composed of an anti-mesothelin Fv fragment fused to a truncated form of Pseudomonas exotoxin A (PE38). It targets mesothelin, a cell surface protein highly expressed in several cancers including mesothelioma, and delivers the toxin to kill tumor cells by irreversibly inhibiting elongation factor-2 and halting protein synthesis[2]. - **Cisplatin** is a platinum-based chemotherapeutic agent that forms DNA crosslinks, leading to apoptosis. - **Pemetrexed** is an antifolate antineoplastic agent that inhibits multiple enzymes involved in folate metabolism and DNA synthesis. The rationale for this combination stems from the potential synergistic effects between targeted immunotoxin therapy (SS1P) and standard chemotherapy (cisplatin plus pemetrexed), aiming to improve outcomes in patients with mesothelin-expressing tumors such as malignant pleural mesothelioma[2][1].
