ST-067 + pembrolizumab is an investigational combination therapy consisting of ST-067, a first-in-class engineered cytokine (decoy-resistant interleukin-18, IL-18), and pembrolizumab, a monoclonal antibody anti-PD-1 checkpoint inhibitor. ST-067 has been specifically engineered to avoid inhibition by IL-18BP, the decoy receptor that neutralizes native IL-18, thus enabling sustained immune activation within the tumor microenvironment. Pembrolizumab blocks the PD-1 immune checkpoint to enhance T cell-mediated anti-tumor responses. The combination is being studied in patients with advanced solid tumors and is intended to overcome resistance to prior immunotherapy. It is currently under evaluation in Phase 1/2 clinical trials for several solid tumor indications, including melanoma, non-small cell lung cancer, renal cell carcinoma, triple-negative breast cancer, squamous cell carcinoma of the head and neck, and MSI-High tumors[1][2][3].
