ST-067 + teclistamab is a combination therapy being developed by Simcha Therapeutics for the treatment of multiple myeloma. ST-067 is an engineered, decoy-resistant interleukin-18 (IL-18) cytokine designed to bypass the inhibitory effects of IL-18 binding protein (IL-18BP), thereby enhancing the activation and expansion of T cells and natural killer (NK) cells. Teclistamab (Tecvayli) is a bispecific T-cell engager (BiTE) that targets B-cell maturation antigen (BCMA) on myeloma cells and CD3 on T cells, facilitating T-cell-mediated lysis of the tumor cells. The combination aims to leverage the immunostimulatory properties of IL-18 to potentiate the anti-tumor activity of the T-cell engager.
