ST-101 is an allogeneic, off-the-shelf hematopoietic stem cell (HSC) therapy developed by Stratus Therapeutics (formerly Garuda Therapeutics) for the treatment of blood and immune disorders. The therapy is generated using the proprietary Stratus Prime platform, which employs the targeted activation of the **PIEZO1 mechanosensitive ion channel** to differentiate human pluripotent stem cells (hPSCs) into transgene-free, long-term reconstituting HSCs and hematopoietic progenitor cells (HPCs). ST-101 is designed to provide a universal, on-demand graft that eliminates the need for human leukocyte antigen (HLA) donor matching. By removing T-cells during the manufacturing process, the therapy aims to achieve durable hematopoietic restoration and rapid immune reconstitution while significantly reducing the risk of graft-versus-host disease (GvHD). Its lead indications include transfusion-dependent beta-thalassemia and bone marrow failure syndromes.
