St. John's wort extract Ze 117 is a standardized dry herbal extract derived from the aerial parts of Hypericum perforatum L., commonly known as St. John’s wort. It is used primarily for the treatment of mild to moderate depressive episodes and has demonstrated non-inferiority to standard antidepressants in clinical studies[2][4]. The mechanism of action involves inhibition of presynaptic reuptake of serotonin, dopamine, and noradrenaline (norepinephrine), resulting in increased synaptic concentrations of these neurotransmitters[2][4][5]. Additionally, chronic exposure leads to downregulation of beta-adrenoceptors similar to tricyclic antidepressants[4]. Unlike many other St. John’s wort extracts, Ze 117 contains low levels of hyperforin (≤1%), which minimizes drug-drug interaction risk by reducing induction effects on cytochrome P450 enzymes and P-glycoprotein transporters[2][6]. The extraction solvent is ethanol (57.9% V/V), with a drug-extract ratio (DER) between 4-7:1[2].
