**St. John's wort + 5-hydroxytryptophan** is a dietary supplement combination used for mood support, primarily targeting mild to moderate depression, anxiety, sleep issues, and stress relief. St. John's wort (*Hypericum perforatum*), containing active ingredients hyperforin and hypericin, inhibits the reuptake of serotonin, dopamine, norepinephrine, and GABA, increasing their availability in the brain similar to SSRIs. 5-Hydroxytryptophan (5-HTP), derived from *Griffonia simplicifolia* seeds, serves as a direct precursor to serotonin, boosting its synthesis to regulate mood, sleep, appetite, and pain. The combination aims to synergistically elevate serotonin levels but carries a high risk of serotonin syndrome and is contraindicated with serotonergic drugs; St. John's wort also induces CYP3A4, interacting with over 800 medications including contraceptives.[1][2][3][4][5]
