STA9090 + dutasteride is an investigational combination therapy studied primarily for castration-resistant prostate cancer. STA9090, also known as ganetespib, is a small molecule inhibitor of heat shock protein 90 (HSP90), a molecular chaperone essential for the stability and function of multiple oncogenic proteins. By inhibiting HSP90, ganetespib promotes the degradation of client proteins involved in cancer cell growth and survival pathways, including those regulating androgen receptor (AR) signaling. Dutasteride is a 5-alpha-reductase inhibitor that blocks the conversion of testosterone to dihydrotestosterone (DHT), thereby reducing androgen levels and suppressing AR activity. The rationale for combining these agents is to achieve greater suppression of AR transcriptional activity than either agent alone, potentially enhancing antitumor efficacy in castration-resistant prostate cancer[1][2].
