Staphylococcal enterotoxins + autologous leukemic cells is an experimental personalized cancer vaccine strategy developed primarily for the treatment of hematologic malignancies like acute myeloid leukemia (AML). The therapy involves harvesting a patient's own (autologous) leukemic cells and treating them ex vivo with staphylococcal enterotoxins (SEs), such as Staphylococcal Enterotoxin B (SEB) or Staphylococcal Enterotoxin A (SEA). These toxins function as superantigens, which are potent immunostimulatory proteins that bypass traditional antigen processing. They bind directly to the Major Histocompatibility Complex class II (MHC II) molecules on the surface of the leukemic cells and simultaneously to the Vβ region of the T-cell receptor (TCR) on T-lymphocytes. This cross-linkage triggers a massive, non-specific activation of T-cells in close proximity to the tumor cells, intended to break immune tolerance and induce a systemic anti-leukemic immune response.
