The staphylococcal polysaccharide conjugate + toxoid vaccine (specifically the SA3Ag candidate) is an investigational multi-antigen vaccine designed to provide active immunization against *Staphylococcus aureus*. The vaccine employs a dual-action strategy to combat infection: it facilitates bacterial clearance and neutralizes key virulence factors. It contains *S. aureus* capsular polysaccharides type 5 and 8 (CP5 and CP8), which are conjugated to a carrier protein (typically CRM197) to convert the T-cell independent carbohydrate antigens into T-cell dependent antigens, thereby inducing high-affinity opsonophagocytic antibodies. Additionally, the vaccine incorporates a recombinant, non-toxic version of the staphylococcal alpha-hemolysin (alpha-toxin or Hla) toxoid. This component elicits neutralizing antibodies that prevent the toxin from forming pores in host cell membranes, which otherwise leads to cell lysis and immune evasion. This combination approach aims to prevent invasive staphylococcal diseases, such as bacteremia and surgical site infections, by targeting both the pathogen's protective capsule and its secreted toxins.
