Overview

This is a **bivalent conjugate vaccine** designed to prevent infections caused by *Staphylococcus aureus*, particularly serotypes 5 and 8, which are associated with the majority of invasive staphylococcal infections in clinical settings. The vaccine consists of **capsular polysaccharides** from *S. aureus* serotype 5 and serotype 8, each chemically conjugated to a protein carrier—commonly a detoxified form of Pseudomonas aeruginosa exotoxin A (as in StaphVAX) or sometimes CRM197, a non-toxic mutant of diphtheria toxin. Conjugation transforms these otherwise poorly immunogenic polysaccharides into antigens that elicit robust, opsonophagocytic, type-specific IgG immune responses, conferring both active and passive protection against bacteremia and other serious infections caused by these serotypes. The vaccine was developed for populations at high risk, such as patients with end-stage renal disease and those on hemodialysis[2][3][5]. Mechanistically, the vaccine induces protective anti-capsular antibodies that mediate killing of the bacteria through immune mechanisms[2][6].
