This experimental combination immunotherapy, developed by Juan C. Ramos, MD at the University of Miami, is designed to treat aggressive relapsed or refractory leukemias. The therapy consists of two primary biological components: autologous leukemic cells that have been ultraviolet (UV)-irradiated and loaded with STING-dependent activators (STAVs), and a vaccine composed of mature autologous dendritic cells. The STAVs serve as potent adjuvants that trigger the Stimulator of Interferon Genes (STING) pathway within the leukemic cells, enhancing their immunogenicity. When these loaded, inactivated cells are administered alongside mature dendritic cells, the dendritic cells process and present leukemia-specific antigens to the patient's T cells. This process is intended to overcome immune evasion and stimulate a robust, systemic anti-leukemic immune response specifically targeting acute myeloid leukemia (AML), acute lymphoblastic leukemia (ALL), and adult T-cell leukemia/lymphoma (ATL).
