SRT + T-DXd is a combination therapy that pairs stereotactic radiotherapy (SRT) with trastuzumab deruxtecan (T-DXd), an antibody-drug conjugate. T-DXd consists of a humanized HER2-targeting antibody (trastuzumab) linked to a topoisomerase I inhibitor payload (deruxtecan). This combination approach is being investigated for treating HER2-positive breast cancer, particularly in cases with metastatic disease. The TENDANCE study has shown that concurrent administration of T-DXd and radiotherapy (including SRT) appears to be safe with manageable toxicity and promising efficacy outcomes.
