STING agonist (TriForm formulation)

**STING agonist (TriForm formulation)** is a preclinical, ultrasound-activated cancer immunotherapy formulation developed by Vesselon. It uses Vesselon's TriForm platform to co-formulate the cyclic dinucleotide STING agonist MSA-1 with Imagent lipid microspheres, self-assembling liposomes, and free active drug in a single injectable product. Ultrasound exposure induces sonoporation and related biophysical effects that increase local tumor delivery of MSA-1, thereby enhancing STING-dependent innate immune activation, cytokine production, and antitumor immune responses. In syngeneic mouse tumor models, this approach increased tumor uptake of intravenously administered MSA-1 by 6.58-fold and produced tumor regressions at doses that were inactive without the Imagent-ultrasound approach.
