Overview

This combination therapy consists of sturlimogene erparepvec (RP2) and tivozanib. Sturlimogene erparepvec is an enhanced oncolytic herpes simplex virus type 1 (HSV-1) designed to selectively replicate in and lyse tumor cells. It is engineered to express a fusogenic protein (GALV-GP R-), human GM-CSF, and an anti-CTLA-4 antibody-like molecule to stimulate a potent systemic anti-tumor immune response. Tivozanib is a selective small-molecule tyrosine kinase inhibitor that targets vascular endothelial growth factor receptors (VEGFR) 1, 2, and 3, thereby inhibiting angiogenesis and tumor growth. The combination is being evaluated by City of Hope Medical Center in Phase 2 clinical trials for patients with metastatic renal cell carcinoma (RCC) who have progressed after receiving immunotherapy with immune checkpoint inhibitors.
