Overview

**Sudapyridine + rifampin** is a combination of two distinct anti-tuberculosis drugs: sudapyridine (WX-081), a novel small molecule diarylpyridine derivative and analog of bedaquiline that inhibits mycobacterial ATP synthase, and rifampin, a well-established rifamycin-class antibiotic that inhibits DNA-dependent RNA polymerase in Mycobacterium tuberculosis. Sudapyridine is under investigation for its potent in vitro and in vivo efficacy against drug-sensitive and drug-resistant strains of tuberculosis, with improved safety and lower potential for QTc prolongation compared to bedaquiline. Rifampin is considered a cornerstone of standard tuberculosis therapy due to its bactericidal activity but is often compromised in resistant cases. This combination is relevant in the research context of treating rifampicin-resistant or multidrug-resistant tuberculosis, as studied in current or recent clinical trials.
