Overview

**sunitinib + bevacizumab + paclitaxel** is an investigational combination therapy that consists of:\n- **Sunitinib**, a small molecule multitargeted receptor tyrosine kinase inhibitor;\n- **Bevacizumab**, a humanized monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A);\n- **Paclitaxel**, a chemotherapeutic agent of the taxane class that disrupts microtubule function.\nThis combination has been explored in clinical trials for advanced cancers, including non-small cell lung cancer and breast cancer, based on the hypothesis that simultaneous inhibition of angiogenesis at multiple levels (VEGF ligand and receptor) plus cytotoxic therapy may yield superior anti-tumor activity. However, clinical studies have reported that this triple regimen is associated with substantial hematologic and non-hematologic toxicity, leading to high rates of dose reductions, interruptions, and discontinuations. Due to poor tolerability and limited efficacy signals, the development of this combination has not advanced beyond early-phase trials and is not standard of care for any indication[1][2][4].
