Overview

**Sunitinib + prednisone** is an oral pharmaceutical combination used primarily in clinical investigation for metastatic castration-resistant prostate cancer (mCRPC), particularly in patients who have progressed after or during docetaxel-based chemotherapy. **Sunitinib** is a small molecule multi-targeted receptor tyrosine kinase inhibitor, blocking vascular endothelial growth factor receptors (VEGFRs), platelet-derived growth factor receptors (PDGFRs), KIT, RET, CSF-1R, and FLT3. **Prednisone** is a synthetic glucocorticoid corticosteroid with anti-inflammatory and immunosuppressive properties, acting via the glucocorticoid receptor. The combination is studied both as second-line therapy (after docetaxel failure) and in combination with docetaxel for first-line treatment. In clinical trials, this combination improved progression-free survival (PFS) but not overall survival in mCRPC. Sunitinib is developed and manufactured by Pfizer[2][3][5].
