Surufatinib + gemcitabine is a combination therapy under investigation for the treatment of various solid tumors, including pancreatic cancer. Surufatinib is an oral, small-molecule tyrosine kinase inhibitor that selectively inhibits vascular endothelial growth factor receptors (VEGFR1–3), fibroblast growth factor receptor 1 (FGFR1), and colony stimulating factor 1 receptor (CSF-1R). This dual mechanism blocks tumor angiogenesis and modulates the tumor immune microenvironment by regulating tumor-associated macrophages[6][1]. Gemcitabine is a nucleoside analog antimetabolite that interferes with DNA synthesis by incorporating into DNA and causing masked chain termination, leading to cell death[8][3]. The combination aims to leverage both antiangiogenic/immune-modulating effects of surufatinib and the cytotoxic activity of gemcitabine. Surufatinib is developed and marketed in China by HUTCHMED under the brand name SULANDA[1].
