Surufatinib + penpulimab is a combination therapy consisting of surufatinib, a small molecule multi-kinase inhibitor, and penpulimab, a humanized IgG1 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor. Surufatinib inhibits vascular endothelial growth factor receptors (VEGFR 1/2/3), fibroblast growth factor receptor 1 (FGFR1), and colony-stimulating factor 1 receptor (CSF-1R), thereby inhibiting angiogenesis and modulating the tumor microenvironment by reducing tumor-associated macrophages. Penpulimab is a PD-1 inhibitor that restores T-cell mediated anti-tumor immune responses. This combination is being investigated for its synergistic potential in treating advanced solid tumors, including osteosarcoma and soft tissue sarcoma, where the anti-angiogenic and immunomodulatory effects of surufatinib may enhance the efficacy of PD-1 blockade.
