Suvemcitug + trifluridine + tipiracil is an investigational combination therapy being developed by Jiangsu Simcere Pharmaceutical for the treatment of refractory metastatic colorectal cancer (mCRC). Suvemcitug (also known as SIM0235) is a humanized monoclonal antibody designed to target and neutralize vascular endothelial growth factor (VEGF), thereby inhibiting the angiogenesis required for tumor growth and metastasis. Trifluridine is a cytotoxic nucleoside analogue that, once phosphorylated, is incorporated into viral or cellular DNA, leading to DNA dysfunction. Tipiracil is a thymidine phosphorylase inhibitor that prevents the degradation of trifluridine, significantly increasing its systemic exposure and efficacy. This combination is currently being evaluated in a Phase Ib/III clinical trial to determine if the addition of an anti-angiogenic agent to the standard-of-care trifluridine/tipiracil regimen can improve progression-free and overall survival in patients who have progressed on prior therapies.
