SY-1365 + carboplatin is an investigational combination therapy consisting of SY-1365, a first-in-class, potent, selective covalent inhibitor of cyclin-dependent kinase 7 (CDK7), and carboplatin, a standard-of-care platinum-based DNA-damaging chemotherapeutic. SY-1365 works by inhibiting CDK7, a kinase essential for both cell cycle progression and transcriptional regulation, resulting in cell cycle arrest and induction of apoptosis particularly in cancers with heightened transcriptional addiction. Combining SY-1365 with carboplatin, which induces DNA damage, has shown synergistic anti-tumor effects in preclinical models of ovarian cancer and is being investigated in early-phase clinical trials for enhanced efficacy in patients with relapsed ovarian cancer.
