Overview

SY-5609 + gemcitabine + nab-paclitaxel is a triple combination regimen under development for cancer treatment, notably advanced or metastatic pancreatic cancer and other solid tumors. SY-5609 is a highly selective, oral, noncovalent inhibitor of cyclin-dependent kinase 7 (CDK7), which arrests the cell cycle and impairs transcription, leading to tumor cell apoptosis[1][3][5]. Gemcitabine is a nucleoside analog that inhibits DNA synthesis, causing cytotoxicity in rapidly dividing cells. Nab-paclitaxel is paclitaxel bound to albumin nanoparticles, enhancing tumor delivery and reducing solvent-related toxicity, and it disrupts microtubule dynamics, leading to cell cycle arrest[4][8]. The addition of SY-5609 to gemcitabine and nab-paclitaxel is being evaluated in Phase 1 clinical trials for enhanced antitumor activity, based on strong preclinical rationale and established efficacy of the doublet in pancreatic cancer[5].
