SYM004 + cisplatin + gemcitabine is a combination regimen under investigation for cancer therapy. SYM004 is a first-in-class synergistic antibody mixture containing two recombinant monoclonal antibodies, futuximab and modotuximab, which bind to different, non-overlapping epitopes of the epidermal growth factor receptor (EGFR). This dual binding induces rapid and efficient internalization and degradation of EGFR on cancer cells, leading to potent inhibition of EGFR signaling pathways. SYM004 has demonstrated activity in preclinical models resistant to other anti-EGFR therapies such as cetuximab by inhibiting downstream signaling including MET, AKT, and MAPK pathways[1][2][3][4][5][6][7]. Cisplatin is a platinum-based chemotherapeutic agent that causes DNA crosslinking and apoptosis. Gemcitabine is a nucleoside analog that inhibits DNA synthesis. The combination aims to maximize tumor cell kill through complementary mechanisms: targeted EGFR blockade with SYM004 plus cytotoxic effects from cisplatin and gemcitabine.
