Overview

SYM024 + SYM021 is a combination immunotherapy regimen under clinical development for advanced solid tumor malignancies. SYM024 is a fully human, effector-function attenuated monoclonal antibody targeting CD73 (also known as ecto-5'-nucleotidase or NT5E), where it blocks enzymatic activity implicated in adenosine-mediated tumor immune evasion. SYM021 is an anti-PD-1 monoclonal antibody that acts as a checkpoint inhibitor. The rationale for their combination is to simultaneously inhibit the adenosine pathway (via CD73 blockade) and restore T-cell anti-tumor immunity (via PD-1 inhibition), potentially overcoming primary and acquired resistance to immune checkpoint therapy. Both agents are developed by Symphogen and tested in combination in phase 1 clinical trials for safety, tolerability, pharmacokinetics, and pharmacodynamics in patients with advanced solid tumors[1][6][4][5].
