SYS6010 + enlonstobart is a combination therapy being developed by CSPC Megalith Biopharmaceutical for the treatment of advanced or metastatic esophageal squamous cell carcinoma (ESCC). SYS6010 is an antibody-drug conjugate (ADC) targeting the epidermal growth factor receptor (EGFR), consisting of an anti-EGFR monoclonal antibody linked to a Dxd analogue payload (JS1) via a cleavable GGFG tetrapeptide linker with a drug-antibody ratio (DAR) of 8. Enlonstobart (SG001) is a recombinant, fully human monoclonal antibody that acts as a PD-1 checkpoint inhibitor. This combination is currently being evaluated in Phase 2/3 clinical trials as a first-line treatment for ESCC, often administered alongside chemotherapy such as 5-fluorouracil or capecitabine. The dual approach aims to leverage the targeted cytotoxicity of the ADC and the immune-potentiating effects of the PD-1 inhibitor.
