T3011 + cobimetinib is a combination investigational cancer therapy consisting of T3011, a genetically engineered oncolytic herpes simplex virus (HSV-1) encoding an anti-PD-1 antibody and interleukin-12 (IL-12), together with cobimetinib, a selective MEK inhibitor. T3011 is administered by intratumoral or intravenous injection and is designed to replicate preferentially in tumor cells, induce direct oncolysis, and locally express both the immune checkpoint inhibitor (anti-PD-1) and proinflammatory cytokine (IL-12) to modulate the tumor microenvironment and promote antitumor immunity. Cobimetinib inhibits the MAPK/extracellular signal-regulated kinase (ERK) pathway, which is frequently activated in cancers, thereby reducing tumor cell proliferation and potentially enhancing immune response. This combination targets both tumor-intrinsic growth pathways and immune evasion mechanisms.
