It combines tabalumab (LY2127399), a fully human anti-BAFF monoclonal antibody, with bortezomib, a proteasome inhibitor. Tabalumab inhibits B-cell activating factor (BAFF), a cytokine crucial for B-cell survival and implicated in the growth and resistance of multiple myeloma cells. Bortezomib blocks the proteasome-mediated degradation of proteins, triggering apoptosis in plasma cells. Preclinical and clinical studies suggest this combination may help overcome BAFF-related resistance and improve myeloma control by acting on both tumor cell survival and bone disease[1][3][5].
