TAC-T cells targeting FAP (IL12 secreting) is an adoptive cell therapy consisting of T-cells engineered with a T-cell Antigen Coupler (TAC) receptor designed to recognize Fibroblast Activation Protein (FAP). FAP is highly expressed on cancer-associated fibroblasts (CAFs) within the tumor microenvironment of various solid tumors. Unlike traditional CAR-T cells, TAC-T cells utilize the endogenous T-cell receptor (TCR) complex to initiate signaling, potentially offering a more controlled and physiological activation. This specific iteration is further engineered to secrete Interleukin-12 (IL-12), a potent pro-inflammatory cytokine intended to remodel the immunosuppressive tumor microenvironment, enhance T-cell persistence, and recruit other immune cells. It is being investigated, often in combination with other TAC-T products, for the treatment of advanced nasopharyngeal carcinoma and other solid tumors.
