Overview

This combination therapy regimen involves Transcatheter Arterial Chemoembolization (TACE) followed by the multi-kinase inhibitor Lenvatinib and a Programmed Cell Death Protein 1 (PD-1) inhibitor, such as Tislelizumab, Sintilimab, or Camrelizumab. It is being investigated by Sun Yat-sen University as a first-line treatment for unresectable hepatocellular carcinoma (HCC) presenting with spontaneous rupture, a potentially fatal complication with a poor prognosis. TACE is used to embolize tumor-feeding arteries and control bleeding while delivering localized chemotherapy (typically Lobaplatin and Pirarubicin). Lenvatinib provides anti-angiogenic and anti-proliferative effects by targeting multiple receptors, and the PD-1 inhibitor component aims to restore the anti-tumor immune response. This triple-modality approach is designed to provide both local and systemic control of the disease.
