Overview

TACE + penpulimab + anlotinib is a combination regimen consisting of transarterial chemoembolization (TACE), the anti-PD-1 monoclonal antibody penpulimab, and the multi-target tyrosine kinase inhibitor anlotinib. - **Transarterial chemoembolization (TACE)** is a locoregional therapy used primarily for unresectable hepatocellular carcinoma (HCC). It involves delivering chemotherapy directly to the liver tumor via its arterial blood supply, followed by embolic agents to block blood flow, causing ischemia and enhancing cytotoxic effects[2]. - **Penpulimab** is a humanized IgG1 monoclonal antibody that targets programmed cell death protein 1 (PD-1), blocking its interaction with PD-L1/PD-L2 and thereby enhancing T-cell mediated antitumor immunity[8][9]. - **Anlotinib** is a small molecule tyrosine kinase inhibitor that inhibits multiple receptors involved in angiogenesis and tumor proliferation, including VEGFR, FGFR, PDGFR, and c-Kit[1][2]. This triple combination aims to synergistically enhance antitumor efficacy by combining local tumor control from TACE with systemic immunotherapy (penpulimab) and antiangiogenic therapy (anlotinib). The regimen has shown promising results in early studies for unresectable hepatocellular carcinoma associated with hepatitis B virus infection[2], but further validation in larger randomized trials is needed.
