Tafasitamab + bendamustine is a combination regimen consisting of **tafasitamab**, an Fc-modified, humanized monoclonal antibody targeting CD19, and **bendamustine**, an alkylating agent. Tafasitamab binds to CD19, a surface glycoprotein broadly expressed in B-cell malignancies, leading to tumor cell apoptosis, enhanced antibody-dependent cellular cytotoxicity (ADCC), and antibody-dependent cellular phagocytosis (ADCP) via Fcγ receptor interaction on effector cells such as NK cells and macrophages[2][5][6]. Bendamustine crosslinks DNA, resulting in cell death through apoptosis, and is commonly used in hematological malignancies. This combination is being investigated as a therapeutic regimen for relapsed/refractory diffuse large B-cell lymphoma (DLBCL), especially in patients who are ineligible for autologous stem cell transplant (ASCT)[5]. As of September 2025, tafasitamab is approved in combination with lenalidomide, not bendamustine, for DLBCL, but the combination with bendamustine is being explored preclinically and in trials for B-cell lymphomas.
