Overview

Tafasitamab + lenalidomide is a combination therapy approved for the treatment of adult patients with relapsed or refractory diffuse large B-cell lymphoma (DLBCL) who are not eligible for autologous stem cell transplant. Tafasitamab is a humanized monoclonal antibody that targets CD19 antigen, a surface antigen on B cells, leading to their destruction through mechanisms such as antibody-dependent cellular cytotoxicity and phagocytosis. Its Fc region has been engineered to enhance binding to Fcγ receptors, further potentiating immune-mediated killing of malignant B cells. Lenalidomide is an immunomodulatory small molecule that stimulates proliferation and activation of natural killer (NK) cells and T-cells, enhances immune synapse formation, and modulates cytokine production in the tumor microenvironment. The combination leverages synergistic effects by both directly targeting malignant B cells and enhancing anti-tumor immune responses[1][5][6][7][8].
