tafasitamab + parsaclisib

Tafasitamab + parsaclisib is an experimental combination therapy currently under investigation for the treatment of relapsed or refractory B-cell malignancies, including non-Hodgkin lymphoma and chronic lymphocytic leukemia. Tafasitamab is a humanized monoclonal antibody targeting CD19, a surface antigen broadly expressed on B-cells; it enhances antibody-dependent cellular cytotoxicity and phagocytosis by activating natural killer cells and macrophages. Parsaclisib is a selective, potent small molecule inhibitor of phosphoinositide 3-kinase delta (PI3Kδ), a kinase involved in B-cell receptor signaling and B-cell survival. Together, the combination is hypothesized to improve antitumor activity by both directly targeting malignant B-cells and disrupting key pro-survival signaling pathways. The combination is being evaluated in the phase 1b/2a topMIND trial for safety, tolerability, pharmacokinetics, and efficacy in adults with relapsed/refractory non-Hodgkin lymphoma or chronic lymphocytic leukemia[3].
