Overview

This combination therapy consists of the PCSK9 inhibitor tafolecimab, the PD-1 inhibitor sintilimab, and chemotherapy (specifically nab-paclitaxel, docetaxel, or gemcitabine). It is being investigated as a second-line treatment for patients with advanced or metastatic driver gene-negative non-small cell lung cancer (NSCLC) who have progressed after first-line immunotherapy. The rationale for adding a PCSK9 inhibitor to immunotherapy in cancer involves the potential role of PCSK9 in regulating MHC-I expression on tumor cells; inhibiting PCSK9 may enhance tumor antigen presentation and improve the efficacy of PD-1 blockade. The regimen is currently being evaluated in a Phase II clinical trial led by Jinghui Wang at the Cancer Hospital of the Chinese Academy of Medical Sciences.
