Drug intelligence / Profile preview
Tafuramycin-A (TF-A) is a seco-cyclopropyl pyrrolo indole analogue and a duocarmycin-SA derivative that acts as a potent DNA alkylating agent. It irreversibly binds to and alkylates DNA, specifically targeting A-T rich regions. TF-A is primarily utilized as a chemical attenuating agent to prepare whole-parasite blood-stage malaria vaccines (such as attenuated *Plasmodium falciparum* parasites), preventing parasite replication while preserving immunogenicity. Additionally, it has been investigated preclinically for its cytotoxic and anticancer properties, particularly against breast cancer.
