Overview

TAK-071 + donepezil is an investigational combination therapy consisting of TAK-071, a small molecule positive allosteric modulator of the muscarinic acetylcholine M1 receptor (M1 PAM), and donepezil, an acetylcholinesterase inhibitor. TAK-071 selectively potentiates M1 receptor activity with low cooperativity, aiming to improve cognitive function with reduced cholinergic side effects. Donepezil increases synaptic acetylcholine levels by inhibiting its breakdown, indirectly activating both nicotinic and muscarinic acetylcholine receptors. Preclinical and phase 1 studies suggest their combination can synergistically improve cognition in models of cholinergic deficit without exacerbating adverse cholinergic effects. The combination is primarily studied for its potential in neurodegenerative diseases with cognitive dysfunction such as Alzheimer's disease and Parkinson's disease[1][2][3].
