TAK-114 is an orally bioavailable small molecule antagonist of the α4β7 integrin, developed by Takeda for the treatment of ulcerative colitis. The α4β7 integrin is a cell surface glycoprotein expressed on a subset of circulating B and T lymphocytes that mediates their trafficking to the gastrointestinal mucosa by binding to mucosal addressin cell adhesion molecule-1 (MAdCAM-1). By inhibiting this interaction, TAK-114 aims to reduce the infiltration of inflammatory cells into the gut wall, similar to the mechanism of the monoclonal antibody vedolizumab, but in an oral formulation. It reached Phase 1 clinical development but was subsequently discontinued from Takeda's pipeline.
