TAK-117 + docetaxel is a combination therapy consisting of TAK-117 (serabelisib), a selective, orally available small molecule inhibitor of phosphoinositide 3-kinase alpha (PI3Kα), and docetaxel, a cytotoxic chemotherapeutic agent in the taxane class. TAK-117 blocks PI3Kα, thereby inhibiting the PI3K/AKT/mTOR signaling pathway, which is implicated in tumor growth, angiogenesis, and resistance mechanisms in cancer[1][2][3][4][7]. Docetaxel stabilizes microtubules and prevents their depolymerization, leading to the inhibition of cellular mitosis and induction of apoptosis in dividing tumor cells. This combination is being investigated for synergistic antitumor activity, primarily in solid tumors and specific cancers such as breast cancer and other solid malignancies.
