TAK-659 + bendamustine is a combination therapy consisting of **TAK-659** (also known as mivavotinib), a small molecule inhibitor of spleen tyrosine kinase (SYK), and **bendamustine**, an alkylating agent with unique mechanisms resulting in DNA damage and induction of the DNA repair pathway. TAK-659 inhibits B-cell receptor (BCR) signaling, proliferation, activation, and chemotaxis of malignant B-cells. Bendamustine impairs tumor cell growth through alkylation and DNA damage. The combination was investigated primarily for treatment of advanced relapsed or refractory non-Hodgkin lymphoma, where the dual targeting of B-cell signaling and DNA integrity aimed to improve antitumor activity and overcome resistance mechanisms[1][2][3][4].
