TAK-981 + avelumab is a combination of **TAK-981 (subasumstat)**, a first-in-class small molecule inhibitor of the SUMO-activating enzyme (blocking the SUMOylation pathway), and **avelumab**, an anti-PD-L1 monoclonal antibody checkpoint inhibitor. TAK-981 blocks cancer cell progression and stimulates anti-tumor immunity through upregulation of interferon signaling, enhancement of immune cell (macrophage, NK, T cell) responses, and suppression of tumor immune evasion. In preclinical and early clinical studies, this combination has shown the ability to modulate the tumor microenvironment toward an immune-activated state—TAK-981 enhancing and prolonging interferon pathway signaling and promoting antigen presentation, while avelumab blocks PD-L1/PD-1-mediated immunosuppression. The combination is under clinical investigation for its anti-tumor activity, especially in oncology indications (e.g., head and neck carcinoma)[2][4].
