Overview

TAK-981 + cetuximab is an investigational drug combination composed of TAK-981, a first-in-class small molecule inhibitor of SUMO-activating enzyme subunit 2 (SAE2), and cetuximab, a chimeric monoclonal antibody targeting the epidermal growth factor receptor (EGFR). TAK-981 acts by inhibiting the post-translational modification SUMOylation, reactivating type I interferon signaling, and thereby promoting both direct anti-tumor effects (such as apoptosis and cell-cycle arrest) and indirect immune-mediated anti-tumor immunity (including increased T cell and NK cell activation) in cancer models[1][3][7][10]. Cetuximab binds to EGFR, blocking downstream proliferative and survival signals, and also induces antibody-dependent cellular cytotoxicity. This combination is being explored for synergistic anti-tumor activity in solid tumors and hematological malignancies, with TAK-981 contributing immunomodulatory and cytotoxic effects, and cetuximab providing EGFR blockade and cytotoxicity via immune effector engagement[1][7].
