Drug intelligence / Profile preview
Talazoparib + enzalutamide is a fixed combination therapy used to treat adults with metastatic castration-resistant prostate cancer (mCRPC) that has spread and harbors mutations in homologous recombination repair (HRR) genes. Talazoparib is a poly (ADP-ribose) polymerase (PARP) inhibitor that blocks the PARP protein, impairing DNA repair in cancer cells and leading to cell death, particularly in tumors with defective DNA repair mechanisms. Enzalutamide is an androgen receptor antagonist that inhibits androgen receptor signaling, blocking hormones from fueling tumor growth even in castration-resistant settings. The combination was approved by the FDA based on improved radiographic progression-free survival and overall survival compared to enzalutamide alone[1][2][3][4][5].
